Dear HS and PK Collegeue Please suggest: Can a PK parameters results can vary when calculated using WinNonlin 3.1 and 5.0.1.? we are geting different AUCt when calculated using two different versions. According to me it sholud not because even in higher version of WinNonlin calculation of AUC will be based on standard trapazoidal formula. Hhm, I've got PCNonlin 1.0 (somewhere on a 3.5" floppy disk), WinNonlin 3.3, 4.0, 5.0.1 on CD-ROMs, 5.1.1 as a ZIP-file, and the current version 5.2 installed. Since it's not possible to keep different versions on the same system, I would suggest to check the calculation according to the 'Getting Started.pdf' (v3.3, probably also v3.1). Does your data set contain missing values? If yes, check that the NCA-Model Options - NCA Settings are the same in both versions. WinNonlin uses 'Linear Trapezoidal (Linear Interpolation)' by default - which I would not recommend; it's better to use 'Linear Trapezoidal (Linear/Log Interpolation)'. ❝ According to me it sholud not because even in higher version of WinNonlin calculation of AUC will be based on standard trapazoidal formula. Yes, this is WinNonlin's default in all versions, but it's possible to change it according to your preferences in 'Tools - Options... - Models - NCA calculation method'. Only if the calculation is set to the same method in both versions, results should be the same. I would also suggest to use a spreadsheet or - even better - a pocket calculator to compute AUC from the one profile you're getting different results in WinNonlin.
