Calculation of power in BABE study (WinNonlin) [Software]

hai,
Can anybody pls tell me, what is the formula for calculating the power in BABE studies? i.e In WinNonlin 5.0.1 at the time of Bioequivalence Analysis the output had the power values for that one what is the formula for calculating power.
pls drop the answer.

 A-posteriori power (WinNonlin)

Dear Raghavendra,

the formula is given in both the printed manual and the online help system of WinNonlin 5.0.1 (bioequivalence wizard section).
Since I'm vacationing, I cannot give you a more detailed answer... :PCchaos:

Anybody Could explain with example how to calculate power?

Because iam getting problem in calculating the probabilities, which are given in the formulas.

 A-posteriori power (WinNonlin)

Dear shekarsunkoju!

❝ […] iam getting problem in calculating the probabilities, which are given in the formulas.

Not even with the formulas, but also the program's result:

Open 'Data22.PWO' in folder 'Examples', and start the BE Wizard
Set the 'Reference value' to 'Capsule', 'Next >'
Drag 'AUClast' to 'Dependent Variables', 'Next >'
Keep defaults in the 'Variance Structure' Window, 'Next >'
Keep defaults in the 'Bioequivalence Options' Window, 'Next >'
In the 'Bioequivalence General Options' Window:
Check the 'ASCII Output' box and change 'Degrees of Freedom' to 'Residual', 'Calculate'

ASCII-Output (shortened):
Final variance parameter estimates:
    Var(Sequence*Subject)    0.0979228
            Var(Residual)    0.0532774
          Intersubject CV    0.320745
          Intrasubject CV    0.233928

    Ratio(%Ref) =   104.6588

Power of ANOVA for Confidence Level 90.00
    Power at 20% = 0.898818

But I get (for 104.6588%, 0.233928) values above WinNonlin’s:
0.9582836 (R-code)
0.95828  (StudySize2.01)
0.9582    (nQuery Advisor 5.0)
For a ratio of 100% I get 0.9906747 – and WinNonlin’s value for a ratio of 93.2701%.
So what the h… is WinNonlin actually calculating here?

BTW, a posteriori power does not exist; see:

Thanks.

Dear Kevan!

❝ Did you means that WinNonlin power calculation can not be trusted?

No; only that:
  • A posteriori power calculations do not make any sense anyway (see above in this thread and also this post).
  • I have no idea (based on formulas given in the manual) how WinNonlin calculates the value. The t-distribution (instead of the noncentral t) is used.
I asked Pharsight’s Support a couple of years ago, but didn’t get a useful response – apart from telling me in a polite way to RTFM.

Discussions on WinNonlin's BE wizard on Pharsight Extranet

Dear Helmut,

Thanks for raising some of these points on the Pharsight Extranet. I've been away this week but I note my inbox has several discussions about how and when to address these in the Phoenix release schedule. There will be a reply on the board shortly so hopefully the above cross reference will help your readers.

Simon.
