For Loo-Riegelman you require the ‘true’ elimination from an iv administration. I guess you have it. However, any PK model (and deconvolution based on it) is purely empirical. There is no relationship to physiology.* Hence, you cannot distinguish between liver first pass and pre-systemic first pass in the gut wall. If you are interested in them, you need PBPK.

Did you bother reading my previous post? You can’t estimate liver first pass with a PK model. If you have – a lot of – money, go for suitable software (SymCyp, GastroPlus). Even then, there is no guarantee that it will work for your drug (I have seen terrific failures).
