 Au sujet de la conservation du plasma frais congelé vierge [Bioanalytics]

je vous prie de me renseigner sur les conditions de stockage des poches de plasma vierge destinées pour la bioanalyse , tout en sachant que lors de ma recherche je suis tombée que sur les bonnes pratiques de manipulation du plasma à usage thérapeutique .

Ma question est la suivante : est ce que le plasma frais congelé utilisé pour surcharger les échantillons pendant la validation doit répondre à des règles de conservation, cycles de congélation /décongélation même s'il est destiné à l'extraction après (pas besoin des constituants de plasma).

Lors de son utilisation j'ai remarqué pour le même lot l'apparitions de nouveaux pics dans les échantillons de blanc matrice qui n'existaient pas avant, malheureusement ne disparaît pas suite à l'extraction et il interfère avec mon pic d’intérêt .s'agit il d'une dégradation de plasma ?

Actuellement mon plasma frais congelé est mis à -80°C

(English summary of the question and of my answer at the end of this message, for the non-French speakers. I'm not sure Helmut encourages the use of languages other than English on the forum).

Une première précision, du fait des termes utilisés (poches de plasma, plasma frais congelé) : j'espère que vous n'utilisez pas du PFC obtenu auprès d'une banque de sang et prélevé sur CPDA. C'est une source de problème énorme en bioanalyse : la proportion d'anticoagulant utilisée est très importante et conduit à une dilution du plasma d'environ 30 % par rapport à un prélèvement sur EDTA ou héparine, généralement utilisé pour les échantillons de l'étude.

Concernant votre question, il y a 2 aspects :
- les conditions de conservation du plasma avant son utilisation ;
- la problématique des cycles de congélation/décongélation.

Pour la température de conservation : je vois la plupart du temps une conservation à -20°C ou -30°C, ce qui est suffisant dans la très grande majorité des cas. La durée de conservation ne doit pas être trop prolongée : on peut avoir une dégradation progressive de certains constituants du plasma, qui peut provoquer des interférences (voir par exemple ce poster décrivant l'apparition progressive d'une interférence dans du plasma de donneurs masculins). C'est peut-être ce qu'il s'est passé pour ce que vous décrivez : apparition de nouveaux pics qui n'existaient pas avant.

En cas de problème pour une méthode donnée : utiliser du plasma le plus frais possible ; une conservation à -80°C pourrait être justifiée.

Pour les cycles de congélation/décongélation : c'est souvent plus problématique que la durée ou la température de conservation. Les échantillons décongelés sont souvent troubles, avec un précipité protéique. La multiplication des cycles de congélation/décongélation amplifie les problèmes. On peut aussi avoir une modification progressive du pH et une oxydation au contact de l'air à chaque ouverture. Les conséquences peuvent être l'apparition d'interférences, des modifications de rendement d'extraction... Voir par exemple cet article* sur les modifications progressives (il y a certainement des articles plus récents sur le sujet si on cherche un peu).

Si vous obtenez du plasma congelé dans des contenants de volume important (par exemple 200 ml) : plutôt que de recongeler le volume non utilisé en une seule fois, il est préférable de l'aliquoter en conteneurs plus petits (par exemple de 10 ml ou 30 ml) et de ne décongeler ensuite que le nombre de tubes strictement nécessaire. On peut ainsi limiter le nombre de cycles. Si vous recevez le plasma frais, non congelé, aliquotez le directement.



The question relates to the storage of blank plasma: recommendations for storage conditions, freeze/thaw cycles (considering that the plasma will be extracted, the protein themselves will not be analysed). Developper bioanalyste mentions that he had interfering peaks appearing in blank plasma, which were not there initially. He stores his blank plasma at -80°C.

First a clarification, due to the terminology used (plasma bags, fresh frozen plasma): I hope you're not using fresh frozen plasma obtained from a blood bank and collected on CPDA. That's a huge source of problems in bioanalysis: there is a large volume of anticoagulant buffer and the blank plasma is diluted by about 30% compared with subject plasma collected on EDTA or heparin.

Regarding your question, there are 2 aspects:
- storage conditions for blank plasma
- freeze/thaw (FT) cycles.

Regarding storage conditions: most of the time I see a storage at -20°C or -30°C, which is usually sufficient. The storage duration should not be too long, as you can have a gradual degradation of some plasma components, which can create interferences (see for instance this poster describing the gradual appearance of an interference in the plasma of male donors). This might be what happened in the case you describe: appearance of peaks which did not exist previously.

In case of problems for a given method, this could justify the storage of blank plasma at -80°C.

Regarding FT cycles: this is often more problematic than the duration or temperature of storage. Thawed samples are often cloudy, with a protein precipitate. Increasing the number of FT cycles amplifies problems. You can also get changes in pH and an oxidisation when opening the container. Consequences can be new interferences, changes in recovery... See also this paper* (you can certainly find more recent papers if searching).

If you get plasma frozen in large volumes (e.g. 200 ml): rather than re-freezing the whole remaining volume in a single container after the first use, is is preferable to aliquot it in smaller containers (e.g. 10 ml, 30 ml), and then only thaw the number of tubes strictly required and not re-freeze again. If you get your plasma fresh: aliquot it before freezing it.

Nope, intentional. That’s how it is used in Vienna (following the style of H.C. Artmann). The entire sentence in Standard German:

Wienerisch is a dialect of Austrian German (which differs* from Standard German). That’s the one I grew up with and still use in daily life. I learned Standard German in primary school. Was tough. No surprise that the artificial unintelligence Google gives up.

BTW, few people master it to speak English with no accent. Native speakers told me that the German accent is reminiscent of the Third Reich, whereas the – more soft – Austrian accent is “charming”. ;-) For me both sound terrible.
