 Matrix effect and Matrix Factor [Bioanalytics]

Dear Member,

Is it require to perform matrix effect and matrix factor both in a validation or only matrix factor is sufficient to prove ion supresion or enhancement.

I'm not sure I understand your question: calculating the matrix factor is one of the possible ways to study matrix effects, and that's indeed the way currently recommended as part of validation. What do you exactly mean with "perform matrix effect and matrix factor both" ?

Dear Sir,

We are performing both of the experiment during the validation as below,

1. Matrix Factor: in this we are comparing responce of aqeous with the reconstitute plasma samples and calculating matrix factor.

2. Matrix effect: we are spiking LQC and HQC in duplicate in same lot (repeated by using 06 different lot of plasma) of plasma and evaluating the same with fresh calibration curve. By this we are monitoring the accuracy of spiked samples.

Now my understanding is if the matrix factor is calculated then there is no need to perform the same experiment under diffrent title and different procedure.

Kindly correct my understanding and explain the correct practice.

Dear Auditor,

If you are calculating matrix factor on 6 different batches of plasma: then the two procedures are indeed equivalent, and doing only one would be sufficient. The White Paper from the 2006 Crystal City meeting recommends to go for matrix factor.

If matrix factor is calculated on only one lot of plasma: then you need to either keep both procedures, or go for 6 lots in matrix factor...
