Hi to all, I have query regarding selection of the matrix for validation, we are planned to conduct study on oral contraceptive on female for this we want to develop analytical method,which biological matrix (Male serum/ female serum) should be taken for development and validation work? Dear Santosh, first you have to demonstrate that your method is selective enough to distinguish the analyte from natural sexual steroidal hormones. I would suggest to go for serum/plasma of females only (just 2% of testosterone than in males' serum/plasma) to avoid troubles. If your study will be performed in premenopausal women (which I would not suggest) mainly check for 17ß-estradiol and estriol, if you are aiming at postmenopausal women check for estrone and 17ß-estradiol. Note: if seen studies using pooled male/female serum/plasma as well, but I would not recommend it.
