 ISTD variation in selectivity experiment [Bioanalytics]

Dear sir,
Is there any specific guidelines for internal standard variation in selectivity experiment, I have processed & injected 6 replicates of LOQ samples, out of 6 replicates in 4 samples ISTD response was observed 1.25 lakhs and rest of 2 samples ISTD responce was observed 3 lakhs, please clarify how to handle this type of situation.

 ISTD variation: matrix effects ?

Dear K.sathis & venkat,

I suppose that you tested 6 different sources of matrix. If you are using LC/MS/MS, you should pay a specific attention to matrix effects investigation, and if possible determine the matrix factor in the same 6 lots of matrix. This could be the explanation for such a variability, and then a source of trouble with your method.
