 Selectivity sequence [Bioanalytics]

Hi,
Can anybody suggest the sequence for selectivity experiment in validation?
Should we run it with a calibration curve?
As the new FDA guidance has given the acceptance in terms of accuracy as well.

And should we evaluate the effect of concomitant medication in selectivity?
