What is the need to go down upto 5 or 7 or 10 half lives while developing a bioanalytical method?

Dear Deepti Maru,

The need to have lower LOQ (5,7 or 10 half lives) should be correlated to the number of half lives upto which sample is collected as per protocol or likely to be collected

If the sampling is carried out upto 6 half lives - For a drug with 12 Hr Half life, the last sampling point will be 72 hours. Under such scenario, LOQ can be upto 7 half lives

This will ensure possibility of minimal BLQ samples in absorption / elimination phase.

It is necessary to have a reproducible LOQ
