 master stock preparation

Dear Ramesh!

❝ for exampler as per the method my nominal concentration of the master

❝ stock is 1000 mcg/ml.

If you don't have anything else in place - describing the allowed accuracy of the weighing procedure - the number 1000 would suggest anything between 999.5 to 1000.4 as acceptable (otherwise you probably should have used the value 1000.0).

❝ if i prepare the stock of concentration 1000.2mcg/ml, is it possible to

❝ adjust the stock concentration of the prepared stock to concentration as

❝ per the method.

There's nothing to adjust for. Use the concentration 'as is'. If your LOQ in validation was (by the chance of an exact weighing) let's say 10 ng/ml; with the new stock solution it's 10.002 ng/mL. So what...

❝ can i do this procedure if no please give me justification for the same.

Which procedure; justification for what?
