Hi everyone, Anyone has experience in modeling pk profile with enterohepatic recirculation? Literature references would be very helpful. Thanks John |

Hi John, Gabrielsson/Weiner (all editions) contains “PK40” in classical WinNonlin-code. I modified it a while ago for po dosing. In my experience the biggest obstacle was finding constraints for the gall bladder emptying time if the reabsorbed fraction was not pronounced. Have you tried using the graphic editor in Phoenix to do this? How do you factor in the bile emptying interval (or the "lagtime" before the bile is released into the gut and subsquently the reabsorption from the GI")? Thanks John |

❝ Nope. I only once translated a two-sequence absorption model (MPH I made a model for biphasic (or mixture) release as well for a drug but this enterohepatic gets me beacause I don't know the amount from the biliary compartment to begin with (brain cramp for now). I will post something later for you to play with when you have time. Thanks John P.S. I am looking into PK40 now. Linda from Pharsight posted it for me after my message on Pharsight's forum. |

Hi, In case anyone else is also looking for the Phoenix code to attempt modelling Enterohepatic Recirculation; here is the direct link to the suggested cross posting.
