Hi John,

Gabrielsson/Weiner (all editions) contains “PK40” in classical WinNonlin-code. I modified it a while ago for po dosing. In my experience the biggest obstacle was finding constraints for the gall bladder emptying time if the reabsorbed fraction was not pronounced. More an art than science.

Question... Have you tried using the graphic editor in Phoenix to do this? How do you factor in the bile emptying interval (or the "lagtime" before the bile is released into the gut and subsquently the reabsorption from the GI")?

Nope. I only once translated a two-sequence absorption model (MPH :-D) from my classical WNL-code to PHX. Took me days… I’m too busy right now to give you the details – I suggest to ask at Pharsight’s Extranet in the meantime. It would be useful if you could upload a project containing data of one profile.

I made a model for biphasic (or mixture) release as well for a drug but this enterohepatic gets me beacause I don't know the amount from the biliary compartment to begin with (brain cramp for now). I will post something later for you to play with when you have time.
