 Bioequivalence conclusion [General Sta­tis­tics]

Some time there are instances in bioequivalence where one of the 90% confidence limit is not falling in the range (80%- 125%), so in this situation can bioequivalence be concluded? And how many PK parameters (AUC, Cmax, tmax) are required to conclude the bioequivalence or can we make conclusion of bioequivalence only on the basis of AUC?

Yes for all BE Study You have to conclude the Bioequivalence. If it is not falling in the range then it (test with reference) will be not bioequivalent

No, you can't give the conclusion on the basis on only AUC.

Right, but what’s primary?
  • US-FDA: Cmax, AUCinf and AUCt; if clinically important, early exposure (AUC truncated at median tmax of the reference)
  • EU-EMA: Cmax, AUCt (no AUCinf!)
  • Canada-HPB/TGD: AUCt and the T/R ratio of Cmax within 80%-125% (no confidence interval); if clinically important, T/R-ratio of AUC truncated at median tmax of the reference (no confidence interval)
  • ...
It is always important to study the guidelines of the target country.
