AUC correction of absorption profiles [Dissolution / BCS / IVIVC]

Dear group,

Say, If I have a BE study data with AUC "100 ng·hr/ml" for treatment A and "90 ng·hr/ml" for treatment B. While generating absorption profiles (wagnor nelson method) for A and B, I get a overlaping profiles.
The question is do I need to correct the profile of B with AUCs ratio (ie., 0.9) for further utility?

Thanks
Ashok

Dear group,
Is it so silly question or none in the group know the answer?
Replies are highly appreciated.

thanks

 AUC correction of absorption profiles

Hi Ashok,
I guess my question first would be what is "further utility"? Are you trying to build some sort of IVIV correlation or ???

I think that a simple 0.9 correction between AUCs may be too approximate; the Wagner-Nelson method assumes that the body acts as a single compartment and then you are adjusting the ?mean or ? individual AUCs.

What about Cmax and Tmax? do you think are they similar enough ? What about your sampling schedule are you confident that this described the curve well? Do you know if the AUC is dose proportional within the range you intend to work with for both treatments?

First clarify the problem you are trying to solve then compare and assess which model is the best to answer that problem.

Good luck
Simon.
