Please read (and take into consideration !) Helmut's comments at the bottom of your post, if you want to continue this discussion !

I can't say - I'm not sure what you call absolute recovery...

Conclusion on what ? What are you trying to compare (overall efficiency vs. LLOQ :-D) ? What will you be using your method for ? What LLOQ will you need ? No need to go to 0.1 ng/ml if the lowest concentration you expect in your study is 5 ng/ml...
