I gave you already a detailed answer in this post. If you are not happy with nonparametric superposition, you will have to go with compartmental modeling. I’m not in the mood to give all the steps (read the manual first – only if you have specific questions ask again). The steps are:
• Choose a compiled pharmacokinetic model (for Exp1.pwo it’s model 11: two compartments, no lag time)
• Select the “best” model based on residual plots and (mainly) the minimum AIC (for Exp1.pwo it’s w=1/y2)
• Create a new workbook, fill in time values in the first column
• Select PK Model 11, select “Simulate data“
• Select the number of doses, dosed amounts and dosing times
• Paste the model parameters from your original fitting
• Done
