Solubility Testing-Shake flask method [Dissolution / BCS / IVIVC]

Dear All

could you please any one clarify the formula to calculate the quantity of drug substance dissolved in each buffer for solubility test using shake flask method, does we have to consider sample concentration/weight for the calculation? hope the solubility of the drug doesn't going to change even addition of extra amount of drug after attaining the equilibrium.

I hope this would be a very old topic and enormous data available in web but i couldn't get the exact formula.:-D:-D
