Recovery vs. overall efficiency [Bioanalytics]

Dear Ohlbe,

Thank you very much for your clarification. Please tell me the overall efficiency and absolute recovery are the same or not!

And now, you please give me a conclusion for the following comparison i.e. Overall efficiency Vs Limit of Quantitation (here I included the LOQ and the instrumentation):

                       CASE I.
Extraction technique : Protein Precipitation Technique with Acetonitrile.
Internal Standard    : Only One Internal Standard was used
                       for both the polarities.
Overall efficiencies : Drug A; LQC: 101%, MQC: 95%, HQC: 90%
                       Drug B; LQC: 36%, MQC: 37%, HQC: 35%
Limit of Quantitation: 0.1 ng/mL for and 1.0 ng/mL
Instrumentation      : LC-MS/MS; API 5000.


                       CASE II.
Extraction technique : Liquid-liquid Extraction Technique with tBME
Internal Standard    : Two Internal Standards (one for +ve ion mode and
                       other for -ve ion mode) were used.
Overall efficiencies : Drug A; LQC: 66%, MQC: 71%, HQC: 72%
                       Drug B; LQC: 71%, MQC: 71%, HQC: 68%
Limit of Quantitation: 1.0 ng/mL for both the drugs
Instrumentation      : LC-MS/MS; API 4000.
