Regarding subject analysis [Bioanalytics]

Dear all,

We are doing subject analysis of Eprosartan mesylate in HPLC UV detector at 230nm wavelength by using Losartan as ISTD. In this Calibration standards and QC samples getting concentrations(Areas), but in subject samples not getting concentrations(Areas)in all samples.

We have checked without ISTD also getting same problem. We have checked instrument conditions and chromatographic conditions.

Previously we did one study. We got good results by using HPLC UV detector with same conditions.

After that we did in LCMSMS by using same extration method, we got good result. What is the reason behind this?
