Comparison of Two Tablet Strengths [Design Issues]

May I have your thoughts on this: should we compare test drug 1 x 500 mg tablet, test drug 2 x 250 mg tablet and reference drug 1 x 500 mg tablet (3-way simple cross-over design), what would be the implication on bioequivalence? The formulation of the 250 mg tablet is not directly proportional to the 500 mg tablet. Thanks.
