WinNonlin comes with an Examples guide have you tried following that?

I would also highly recommend you plan to upgrade to the current version of WinNonlin, Phoenix WinNonlin 6.3 as soon as possible; it was released at the end of March 2012.

The next release is planned for q1/q2 2013, but will primarily focus on Trial Simulator integration.
