Dear All, I have a few questions regarding sparse sampling designs and SE AUC estimation that I hope you can help clarify. The help page for the `auc` function in the PK R package states: "The AUC (from 0 to the last time point) is calculated using the linear trapezoidal rule on the arithmetic means at the different time points." I suppose SE is calculated taking it into account.

Dear Martin, Thank you for your detailed answer and the useful references! I have a follow-up question regarding the PK R package. According to the documentation states: HL and Vss are functions of the MRT and hence they will not be valid under oral administration. Could you please clarify your position on this? In particular: - Why doesn’t the package output Lambda_z directly, given that it’s estimated to compute AUCINF? - Why is HL calculated from MRT rather than being derived as a function of Lambda_z, especially since these parameters are not valid for oral administration?
