Dear All! The whole PK/PD issue is quite new to me due to the fact that I just graduated from university and biostatistics is a very small part of the curriculum (if it is a part of it at all). Since absolute bioavailability is defined as the dose-adjusted ratio of the geometric mean of AUCs following non-intravenous administration and the geometric mean of AUCs following intravenous bolus or infusion administration, is it valid to draw the conclusion of expecting identical/similar AUCs, whether following intravenous bolus or infusion administration? Only if the elimination is mainly by metabolization you may see in some (rare) cases after a – reasonably slow – infusion higher AUCs than after a fast bolus.
